Understanding polycystic kidney disease:

a systems biology approach


For nearly a century clinicians have known the etiology of PKD, an inherited condition that leads to the progressive loss of renal function in association with cysts in the kidneys, liver and pancreas. Mutated DNA passed relentlessly within families from parent to child is the sole cause. Autosomal dominant polycystic kidney disease (ADPKD), the most common inherited cystic disorder afflicting more than 6,000,000 people worldwide, is transmitted by abnormal genes on either chromosome 16 or 4. For reasons that defy understanding, this disease was abjectly ignored by clinician-scientists and laboratory-based investigators until the last quarter of the 20th century. Something about fluid-filled sacs in the kidneys just did not excite the imagination of scientists or open the pocketbooks of funding agencies.
